The wife and I are heading from San Marcos to Destin to visit some family.
As I make my way along I-10, Are there any stops that I should make?
This is mostly directed to the Cajuns and Jags (since I will be passing through Mobile and Lafayette), but others feel free to chime in.

Look, there are a million places worthy of visiting in the Acadiana Area, but you'll have to leave the Interstate to get to them. I understand if that is not possible.
However, there are a couple of places located on the Interstate that are worth visiting to get that "Cajun" experience.
http://donsspecialtymeats.com/
This is in Scott, a few miles before the Lafayette exits on the Interstate. They are a local favorite and have my favorite cracklins. Stop and get an order of them to snack on for your trip. Plate lunches are exceptional.

Fezzo's is also in Scott next to Don's., but it regularly voted a favorite Cajun Restaurant for all of Acadiana.

We ate at Blue Dog last time we came through. I had Cajun Enchiladas. They were great

Hey, if you want a more upscale place, try this place:
http://www.jolieslouisianabistro.com/
It is located a few blocks from Blue Dog. My wife and I went there for our anniversary and were absolutely blown away. VERY good restaurant.

Mobile's got awesome seafood, and crab claws in particular. Ed's Seafood Shed is among my favorite of the Causeway restaurants (just east of town on Mobile Bay).
Other Mobile suggestions:
True Midtown Kitchen (Dauphin St.)
Fuego Coastal Mexican Eatery
Hungry Owl (featured on Man vs. Food)

You should stop by the USA campus if you get a chance, too. It looks great now that a lot of the construction is nearing completion. Maybe take a gander at Stanky and look in the Mitchell Center (bball arena). It wouldn't be too terribly out of the way from the interstate.

And the best barbecue in town -- The Brick Pit -- is only a couple of blocks away from campus. Brick Pit was also on Man vs. Food.

The old, orginal Dewey Destin, and Old Bay Steamers just before the bridge to Ft. Walton Beach.
Wife and I may be down there this weekend, as well.

I would not spend much time in either Lafayette or Mobile. If you have time, the Pensacola Naval Air Station (home of the Blue Angels) has an aviation museum complete with Cinemax Theater that is the best you will find outside the Smithsonian. I highly recommend it.

Which may explain why Featherhead is from Monroe.
Seriously, Lafayette and Mobile are the two current SBC cities with the most to offer as far as food and fun is concerned. He is right about the Naval Aviation museum in P'cola, though. It is outstanding.
Not AS good, but still outstanding, and about 3 hours out of the way is the Army Aviation museum at Ft. Rucker, AL, in Enterprise.

RE: Roadtrip recommendations
With the "100 Years of Naval Flight" celebration going on right now, there is a ton of cool stuff going on at the Pensacola NAS Museum.
And afterwards you can travel the scenic route to Destin on old 98. See the new Wahoo stadium, historic downtown Pcola, Pensacola Beach. Much nicer than the Interstate.
Just be careful in Gulf Breeze just over the 3 Mile Bridge. Speed trap.
